Scara and Duff are an interesting pair just to listen to their conversation. Dr. Duff will give you a small quest and Professor (her first name, not her title) Scara can actually be a part of the main quest in certain circumstances.

The bases are part of the baseball analogy. If you don't know about American baseball:
The baseball field is also known as a baseball diamond (hence, Diamond City) because of its shape - a square tilted to stand on one of its points (90 degree angles). That point is known as home plate and the other points in counterclockwise order are first, second and third bases. Sometimes a Diamond City Security guard will tell you to "run the bases" - i.e., touch each base in sequence - first, second, third, home. If you do this AND you don't have any mods loaded (hah!) you can get a Steam achievement for Fallout 4.
