Grelod the Kind is the elderly sadistic headmistress of the Honorhall Orphanage in Riften. She beats the children and refuses to allow their adoption. During the quest Innocence Lost, Aventus Aretino, an escapee from the orphanage, is attempting to contact the Dark Brotherhood to arrange for her assassination.
Anise is an elderly conjurer living in Anise's Cabin. She appears to be a lonely alchemist, with several ingredients, some uncommon, lying around her home. Anise will pretend to be a kind, elderly woman if you attempt to speak to her before you learn her secret. Trying to converse with her will have her talk about how difficult life is for her on her own, how she appreciates your company, and how she isn't anyone of importance.
Silvia is an elderly resident of Darklight Tower. She seeks to become a hagraven, a desire that her daughter Illia doesn't share. She is a key figure in the quest Repentance. Illia is an Imperial elemental mage who lives in the Darklight Tower along with her mother, witches, and hagravens. Her mother plans to become a hagraven, but she needs a human sacrifice for this. Illia wants to stop her mother by killing her.

* Who is Helgi?
Helgi was a Nord child from Morthal. She was the daughter of Hroggar. She and her mother died in a fire that destroyed their house. In Norse, Helgi is a male name; the feminine form of it is Helga. Her mother was Helga, although this name is commonly interchanged with Helgi, often representing a bond between friends.

When asked about Grelod, he will respond, "Miss Grelod is... well she's a terrible old crone. No person could be that cruel. I think she's part hagraven. She only lets us out in the yard once. In the morning. And she just stands there watching us. Like I said. Hagraven."

Thank you for following my advice, I fear if you had done otherwise, you might not have escaped Grelod's recent outburst. I have never seen her that angry before, if I'm honest with you it scared me to see her treat you that way.
I believe it will be safer for you in your old home in Windhelm. Please see the carriage fare as a gift from me to you. Rest assured there is no need to pay me back at all.
Brynjolf was recently kind enough to lend me a book about shadows, which taught me a few tips to make some extra money.

Silvia & Grelod my dears, I trust you are well. Patience is all I ask, we shall have power very soon! I have dispatched word to Helgi. If she is wise she will leave that useless Hroggar and help us complete our Darklight coven. Prepare Francois, Hroar and Samuel for their introduction to Hircine. Runa will make an excellent addition to our ranks.
Constance must be kept as busy as possible with cooking and cleaning, so that she is available at the very moment we are ready to proceed to the third phase, execution.

The premise of 'The Hagraven Conspiracy' is that Grelod, Anise & Silvia are collaborating to build a new coven of witches and Hagravens, and they will do so in three stages:
1. Research (Anise's duties)
Planning and leading the development of the coven
2. Acquisition (Grelod's duties)
Collecting potential recruits at Honorhall Orphanage
3. Execution (Silvia's duties)
Performing human sacrfices at Darklight Tower
So by killing Grelod, you are not necessarily performing an evil act, you are actually saving the orphans from a fate worse than death. If you choose not to kill Grelod, the orphans will be turned into wolf guardians (early stages of lycanthropy) & pre-hagraven witches.
