It's not necessary, I change the graphics and in no way is FSR disabled, but if you want to make sure it's saved you can mark it, a future game update and/or even a madness in Unreal can reset the settings in some way
I've never seen a game that allowed you to activate an AMD frame gen and a different NVIDIA DLSS at the same time. As far as I know, they're not compatible. You may have a bug if you use it like this, or it may not even activate the AMD frame gen even if it's marked because of the incompatibility.
